摘要
目的 :观察丁酸钠 (sodiumbutyrate ,NaB)对体外培养的卵巢癌细胞 3AO生长的影响 ,初步探讨其作用机制。方法 :以人卵巢上皮癌细胞株3AO为肿瘤模型 ,采用不同浓度的NaB对其进行干预 ,MTT比色法测定NaB对 3AO细胞增殖活性的影响 ,进一步通过光镜、电镜观察 3AO细胞的形态学改变 ,进行DNA降解分析 ,并采用流式细胞技术分析细胞周期动力学变化。结果 :NaB作用下 3AO细胞的增殖呈时间剂量依赖模式受到抑制 ,一定浓度的NaB作用一定时间 ,可诱导 3AO细胞发生凋亡 ,光镜和电镜下可见典型的凋亡细胞形态学改变 ,DNA降解分析中出现典型的梯形条带 ,流式细胞技术分析发现NaB作用首先引起细胞周期分布的改变 ,随剂量的加大和用药时间的延长 ,凋亡率逐渐升高。结论 :NaB通过诱导凋亡而抑制体外培养的 3AO细胞生长 ,其机制可能与细胞G1期阻滞有关。
OBJECTIVE:To investigate the growth inhibition effect of sodium butyrate (NaB) on ovarian carcinoma cells 3AO and explore its mechanism.METHODS:3AO cells were cultured with graded concentrations of NaB.The proliferation activities of them were measured by MTT colormetric assay.The characters of apoptosis were assessed by light and electron microscopy,DNA ladder degradation and flow cytometric analysis.RESULTS:The result of MTT colormetric assay showed that the proliferation of 3AO cells was inhibited by NaB in a dose time dependent fashion.The morphologic changes were found by light and electron microscope,the characteristic DNA ladder indicated that apoptosis occurred in the procession.Flow cytometric analysis showed that after using NaB,cell cycle distribution changed at first and as the time prolonged apoptotic peak went higher.CONCLUSIONS:NaB inhibits proliferation of 3AO cells by inducing apoptosis and G 1 phase arrest may be involved in the mechanism.
