摘要
目的 :探讨益气补肾药方延缓衰老的免疫学机制。方法 :以老龄小鼠为衰老模型 ,用凝胶迁移率 (EMSA)法 ,观察益气、补肾和益气补肾药方对抗CD3抗体诱导的老龄小鼠T细胞中NF κB活性的影响。用Westernblot分析该药方对NF κB两个亚基 (p65和p50 )的表达。结果 :老龄小鼠T细胞中NF κB的活性低于幼龄小鼠 ,3种药方均可不同程度地提高老龄小鼠T细胞中NF κB的活性 ;而对p65和p50的表达没有明显影响。结论 :益气补肾药方可通过提高T细胞中NF κB的活性 ,改善因衰老而导致的免疫功能紊乱。
AIM: The aim of the present study was to explore the immunologic mechan ism of delaying senescence by Strengthening Vital Energy(SVE), Tonifying Kidney (TK) and combined TK and SVE. METHODS: Mice of 20 months were us ed as senescence model. The effects of the prescriptions on anti- CD3 antibody -induced NF-κB activity and the expression of NF-κB in T cells from aged mi ce were analyzed by electrophoretic mobility shift assay (EMSA) and Western bl ot, respectively. RESULTS: NF-κB activity in anti-CD3 antibody-induced T cells from the aged mice was lower than that from young ones. The three prescnptions raised the activity of NF-κB in the T cells from the aged mice to certain ex tent. Combined TK and SVE had no influence on p65 and p50 subunits expressions. CONCLUSION: The increased NF-κB activ ity may be one of mechanisms underlying the improvement of immune response in a ged mice by Chinese medicines.
