摘要
目的 探讨c erbB2和p5 3基因扩增和表达在口腔鳞癌 (OSCC)发生发展中的作用。 方法 用原位杂交和免疫组织化学技术与图像分析相结合的方法 ,对 30例口腔鳞癌和 5例正常口腔粘膜中c erbB2和p5 3两种基因的扩增、mRNA表达和蛋白表达状况进行了定性、定位、定量研究。 结果 OSCC中c erbB2和p5 3的扩增阳性率分别为 4 6 6 7%和 4 0 % ;而c erbB2和p5 3在OSCC中的mRNA表达率分别是 70 %和 80 % ,蛋白表达率分别是4 6 6 7%和 6 3 33%。这两种基因的扩增、mRNA表达和p5 3蛋白表达在各种性状差异之间均没有显著性差异 (P >0 0 5 ) ;erbB2蛋白表达在不同部位的口腔鳞癌中具有显著性差异 (P <0 0 2 )。 结论 c erbB2和p5 3两种基因的扩增和过表达在OSCC发生发展中都具有重要作用 ,c erbB2蛋白表达增高在腭癌和口底癌的形成过程中具有更重要的意义。
Objective To study the function of the amplification and expression of c erbB2 and p53 in the formation and development of oral squamous cell carcinomas (OSCC). Methods Using in situ hybridization and immunohistochemical technique combined with image analysis, the amplification, the expression of mRNA and protein of c erbB2 and p53 were studied in 30 cases of OSCC and 5 normal oral mucosa tissues on the nature, location and quantity. Results The amplification rates of c erbB2 and p53 were 46 67% and 40% in OSCC respectively; The mRNA expression rates of c erbB2 and p53 were 70% and 80% in proper order, and their protein expression rates were 46 67% and 63 33% in turn. The statistics analysis found that the amplification and mRNA expression of c erbB2 and p53 had no significant difference between distinct sex, tumorous position, malignant classification, invasiveness or metastasis and relapse respectively, nor had the protein expression of p53( P >0 05); The protein expression of c erbB2 had a significant difference among different tumor position( P =0 0123), but had no statistical difference in other properties ( P >0 05); The protein expression of erbB2 in carcinomas of palate and floor of mouth were obviously higher than the expression in lip cancer and gingival carcinoma( P <0 05). There was no significant correlation between the two genes in amplification( r =0 0325, P =0 8647), mRNA expression( r =0 0671, P =0 7248) and protein expression( r =0 0036, P =0 9850), nor were between amplification and mRNA expression and between mRNA expression and protein expression of every gene( P >0 05).Conclusion The amplification and overexpression of c erbB2 and p53 play important roles in the formation and development of OSCC, the overexpression of c erbB2 protein has more important significance in tumorigenesis of carcinomas of palate and floor of mouth; Not only transcriptional regulation but also translational modulation exists on the expression of c erbB2 and p53.
