摘要 目的 探讨P19细胞向神经元定向分化发育的可能性。 方法 经维甲酸 (RA)诱导 4d的P19细胞按神经细胞培养方法培养 12d ,分别用NSE、MAP 2和TH进行免疫组织化学染色 ,用AchE进行组织化学染色 ,鉴定分化细胞的特征。 结果 经RA诱导的P19细胞培养 12d ,大多数细胞转变为类似于神经元的形态 ,长出突起并相互联络成网。经NSE免疫组织化学显示 ,12d培养物中神经元约占 89%。P19诱导的神经元培养 12d ,神经元突起经MAP 2染色呈阳性反应。TH免疫组织化学显示 ,神经元中TH染色阳性神经元约占 0 8%。组织化学显示 ,神经元中AchE染色阳性神经元约占 71 0 9%。 Objective To investigate the possibility of induced differentiation of P19 cells into neurons. Methods P19 cells were exposed to retinoic acid(RA) for 4?d and cultured in neural medium for another 12?d.Neuron specific enolase(NSE),microtubule associated protein 2(MAP 2),tyrosine hydroxylase(TH)and acetylcholinesterase(AchE)were detected by immunohistochemistry and histochemistry to identify the properties of the differentiated cells. Results Most of RA induced P19 cells differentiated into neuron like cells with the formation of neurite networks after cultured for 12?d in vitro. Approximately 89% of the cells showed positive immunostaining for the NSE,0 8% of the neurons contain TH,71 09% of the neurons contain AchE.Conclusion\ P19 cells can differentiate into neurons in high frequency with RA induction in vitro .\;[
