摘要
目的 探讨幽门螺杆菌对肝细胞系HepG2增殖与凋亡的影响。 方法 将不同浓度的cagA(+)Hp和cagA(- )Hp与HepG2共同培养 ,应用细胞计数 ,Hoechst 332 5 8细胞核荧光染色等技术 ,观察Hp菌液对体外培养HepG2细胞的增殖与凋亡的影响。结果 1 HepG2数随着cagA(+)Hp菌液浓度的增大和作用时间的延长而减少 ;2 HepG2凋亡率随着cagA(+)Hp菌液浓度的增大而升高。 3 未发现cagA(- )Hp对HepG2生长增殖与凋亡的影响。 结论 cagA(+)Hp对HepG2细胞具有明显的细胞毒作用 ,cagA(+)Hp抑制HepG2增殖 ,诱导凋亡 ,其效应与Hp菌液的浓度和作用时间呈明显正相关。
Aim To explore the effect of Hp on proliferation and apoptosis of HepG2 cell in vitro Methods HepG2 was co cultured with Hp at different concentrations Proliferation and apoptosis of HepG2 were studied by cell count and Hoechst 33258 staining technigues Results 1 HepG2 cells were decreased with the increasing concentrations of cagA(+)Hp and the prolongation of culture time;2 Apoptosis rate of HepG2 were increased with the increasing concentrations of cagA(+)Hp and the prolongation of culture time;3 No influence was found in cagA( )Hp on the effect of proliferation and apoptosis of HepG2 Conclusions cagA(+)Hp has cytotoxicity on HepG2 by inhibition of proliferation and induction of apotosis of HepG2,and the effect is positive related to the concentration and culture time of cagA(+) Hp
