摘要
目的 研究喜树碱对舌鳞癌Tca8113细胞增殖和凋亡的作用。方法 将不同浓度的喜树碱分别作用于Tca8113细胞24、48、72h。分别用MTF法、光镜和电镜观察,流式细胞术及琼脂糖凝胶电泳法检测细胞增殖和凋亡状况。结果 喜树碱对Tca8113细胞的增殖有明显的抑制作用,且在一定的浓度范围内呈时间和浓度依赖性。最佳浓度范围为0.016~8μg/mL。镜下可见典型的凋亡细胞形态学特征。流式细胞术检测,喜树碱浓度≥1μg/mL,作用48h,可引起细胞凋亡,细胞阻滞于S期。琼脂糖凝胶电泳呈凋亡特征性梯状带。结论 喜树碱对Tca8113细胞有增殖抑制和诱导凋亡作用,其抗肿瘤机制与诱导凋亡有关。
Objective To study the effects of camptothecin (CPT) on proliferation and apoptosis of human tongue cancer Tca8113 cells. Methods Tca8113 were incubated with CPT at different concentrations for 24,48 and 72 h respectively.The effects of CPT on cells proliferation were measured by MTT assay and cell apoptosis by electron rni-croscopy,flow cytometry and agarose gel electrophoresis. Results CPT inhibited the proliferation of Tca8113 cells in a time-and dose-dependent manner.The maximum effective concentration for Tca8113 ranged from 0.016μg/mL to 8μg/ mL.Typical morphological features of apoptotic cells were detected by electron microscopy.Flow cytometry test showed that CPT with concentrations higher than 1μg/mL could induce apoptosis in Tca8113 cells after 48 h treatment.And the Tca8113 cell cycle was arrested in S phase.In addition.agarose gel electrophoresis test showed characteristic DNA ladder. Conclusion CPT could inhibit proliferation and induce apoptosis in Tca8113 and may play a role in treatment of squa-mous carcinoma.
