摘要
目的 探讨三氧化二砷 (As2 O3 )对肝母细胞瘤细胞系 (HepG2 )生物学行为、核基质及其相关蛋白肝细胞核因子 4(HNF4)之影响。方法 1μmol/LAs2 O3 处理HepG2细胞 5d ,倒置显微镜观察细胞形态 ;提取并纯化核基质蛋白 ,应用双向电泳分析核基质蛋白的改变 ;蛋白杂交(Western)分析HNF4的表达。结果 与未处理对照组相比 ,1μmol/LAs2 O3 可诱导HepG2细胞分化 ,改变HepG2细胞核基质蛋白的分布 ,下调HNF4在核基质蛋白中的表达。 结论 低浓度As2 O3可诱导HepG2细胞分化的过程并改变核基质蛋白的分布。核基质相关蛋白HNF4是As2 O3 发挥其抗癌作用的靶蛋白。
Objective To investigation the effect of low concentration (1 umol/L) arsenic trioxide (As 2O 3) on nuclear matrix proteins as well as on the expression of nuclear matrix associated protein hepatocyte nuclear factor 4 (HNF4) in human hepatoblastoma cell line (HepG2).Methods Using high resolution two dimensional polyacrylamide gel electrophoresis,the nuclear matrix proteins of HepG2 with low concentration As 2O 3 treatment were compared with control.Western blotting was used to detect the expression of HNF4 in nuclear matrix proteins.The morphological change in HepG2 cells were observed under phase contrast microscopy.Results Low concentration of 1 μmol/L As 2O 3 can induce the differentiation of HepG2 cells and alternate the distribution of nuclear matrix proteins.This concentration of As 2O 3 can down regulate the expression of HNF4 too.Conclusion As 2O 3 can induce differentiation of HepG2 cells involved by HNF4 and alternate the distribution of nuclear matrix proteins in HepG2 cells.
