摘要
目的 :探讨羊Ⅲ型前胶原 (gPCⅢ )放免法在大鼠肝纤维化模型中的应用价值。 方法 :用四氯化碳吸入法和猪血清免疫法建立大鼠肝纤维化模型。每周采血用放免法测血清PCⅢ含量。取肝脏标本经组织学观察确定肝纤维化级别。用双盲法比较肝病理纤维化分级与放免法血清PCⅢ结果的相关性。结果 :大鼠肝病理纤维化分期与血清PCⅢ水平相关趋势明显。结论 :gPCⅢ放免法可用于动态监测大鼠血清PCⅢ变化 。
Objective To explore the usefulness of radioimmunoassay with goat procollagen typeⅢ (gPCⅢ) reagent for evaluation of degree of hepatic fibrosis in rat models. Methods Rat hepatic fibrosis models were established by daily inhalation of carbon tetrachloride gas or immunization with pig serum injection two times per week for the whole experiment period. Two model animals (one from each group) were sacrificed weekly for six weeks, taking blood samples for RIA with PCⅢ reagent and liver specimens for histologic examination of the degree fibrosis. Blood samples for RIA were also drawn from all surviving animals weekly. Results Significant elevation of serum PCⅢ levels could be noted at as early as d8 in the CCl 4 group and d15 in the pig serum group with steady increase up to the end of experiment (d36). Degree of hepatic fibrosis also correspondingly progressed. Correlation tendency between degree of fibrosis and serum PCⅢ levels was significant in both groups. Conclusion Measurement of rat model serum PCⅢ codcentration by RIA with PCⅢ reagent is very sensitive and more ideal than that with human PCⅢ reagent.
