摘要
伊贝母鳞茎切块培养在附加不同浓度的2.4—D 和 KT 的 MS 固体培养基上,对其愈伤组织细胞染色体的变异进行了观察和研究.结果表明,在 KT 浓度一定的情况下(0.1毫克/升),随着2.4—D 浓度的增加,染色体变异频率越高;在2.4—D 浓度一定的情状下(1.0毫克/升),随着KT 浓度的增加,染色体变异频率越高.当2.4—D 和 KT 浓度分别为分别1.0毫克/升和0.5毫克/升时,变异频率达到最高.2.4—D 和 KT 浓度都为0.1毫克/升时,变异频率最低.根据实验结果,对2.4—和 KT 引起染色体变异的原因及2.4—D 与愈伤组织形成的关系进行了讨论.
The cutting pieces of bulb of F.pallidiflora Schrenk were culturedon MS media supplemented with different concentration f 2,4-D anb KTChromosome variation in calli cells were observed and studied.The resultshowed that when KT concentration is the same(0.1mg/1),frequency ofchromosome variation is higher and higher to follow increase of 2,4-Dconcentration;when 2,4-D concentration is the same(1.0mg/l),frequencyot chromosome variation is higher and higher to follow iucrease of KT,concentration.When concentration of 2,4-D and KT are 1.0mg/l and0.5mg/lrespectively,frequency of variation is the highest.When both ofthem are 0.1mg/l,frequency of variation is the lowest.In accordancewith results of our experiments,the possible reason for chromosome varia-tion and relations between 2.4-D and callus formattion were discussed.
