乳腺癌组织中HER2与TOPOⅡα基因表达的检测及相关性分析 被引量:1

摘要 目的 检测人表皮生长因子受体Ⅱ (HER2 /neu)与拓扑异构酶 (TOPOⅡα)在乳腺癌组织中的表达 ,探讨二者的相关关系及在乳腺癌临床化疗中的潜在价值。方法 采用RT -PCR和免疫组织化学方法分别检测 2 4例乳腺癌组织和相应的癌旁组织的HER2、TOPOⅡα的转录和蛋白质水平的表达。结果 RT -PCR结果显示 ,乳腺癌组织中HER2和TOPOⅡα阳性率分别是 2 9% ( 7/2 4)和 92 % ( 2 2 / 2 4) ,明显高于癌旁组织 [4 % ( 1/ 2 4)和 2 1% ( 5 / 2 4) ,P <0 0 0 1] ;免疫组化检测癌组织发现HER2和TOPOⅡα的表达率分别为 3 0 % ( 6/ 2 0 )和 94% ( 16/ 17) ,与RT -PCR结果一致 ,且 6例HER2表达阳性的组织中TOPOⅡα表达均为阳性 ,二者的表达相关 ,R =0 5 2 4,P <0 0 0 1。结论 HER2和TOPOⅡα在乳腺癌组织比癌旁组织有明显增加的表达率 ,且两基因的表达相关 ,提示两基因表达的上调在乳腺癌的发生及恶性表型维持中可能起一定作用。 Objective To detect the expression of HER2 and TOPOⅡα in human breast cancer to explore the relationship between them and the potential value in the chemotherapy of breast cancer. Method 24 breast cancer tissues and paired normal tissues were investigated. RT-PCR and imumohistochemical staining were used to detect the expressions of HER2 and TOPOⅡα at the transcription and protein level respectively. Results HER2 mRNA expressed positively in 29% (7/24) breast cancer tissues vs 4% (1/24) in paired normal tissues. The positive expression of TOPOⅡα mRNA in breast cancer tissues and paired normal tissues were 92% (22/24) and 21%(5/24) respectively. The result of the immunohistochemical staining showed that the positive rates of HER2 and TOPOⅡα in breast cancer tissues were 30%(6/20) and 94%(16/17) respectively, which was consistent with RT-PCR. The 6 cancer samples with HER2 positive expression coexpressed TOPOⅡα. The expression of these two proteins was correlated(R=0.524,P<0.01). Conclusion The expression of HER2 and TOPOⅡα significantly increased in breast cancer tissues compared with the paired normal tissues, suggesting upregulation of the two genes expression may play a role in the pathogenesis of human breast cancer.
