摘要
枸杞叶片接种于附加6—BA0.5mg/1+NAA0.5mg/l 的 MS 培养基上,首先切口处的叶肉细胞被起动,相继是表皮下2~5层叶肉细胞被起动进行脱分化形成大量愈伤组织.在愈伤组织中主要是外缘表层细胞继续活跃地进行细胞分裂,从而向外突起,分化形成芽原基.在这一全过程中细胞的DNA 合成是先于细胞形态转变,因此 DNA 合成的动态可以作为细胞形态转变的起始信号以及进行定位和跟踪研究.
The cutting pieces of leafves in Lycium barbarum L were cultured onMS media supplemented 0.5mg/16-BA and 0.5mg/NAA.The buds weredifferentiated after 25 days in culture and the regenerated plants formedafter 40 days.Cytohistological observations indicated that the dedifferentia-tion occurred primarily in the mesophyll cells near the cutting sites,thenthe middle mesophyll cells.After active dividing,a large number of calliwere obtained.Bud primordia were differentiated from calli dueto divisionsof external layer cells in calli.During this course the celellular DNAsynthesis was earlier than morphological changes.Dynamics of DNA synth-esis may be considered as an initial signal of cell morphological changes.It is also important to make localization of DNA and tracking study.
