用PCR技术同时检测微量血痕的种属来源及其DNA多态性

摘要 应用聚合酶链反应技术扩增血痕DNA,根据扩增产物中有无aPoB基因的数量可变重复序列(VNTR)及其扩增片段的大小即可同时确定其种属特异性和DNA的多态性。实验结果显示人血痕可见扩增带并具有多态性,而猴猪狗猫鸡血痕无扩增带。 How to use trace bloodstain to obtain more information is one of the most important methods in individual identification in forensic biology. Using the polymerase chain reaction (PCR) with apoB oligonucleotide primers simultaneously determining both the species origin and DNA polymorphisims in a same trace bloodstain are reported. The results demonstrate that the specific DNA fragments and polymorphisims were observed with human bloodstain DNA as the template, but not with the DNA from monkey, pig, dog, cat, and chicken bloodstains. As a result, we found a potentially reliable method for species and individual identification of a trace bloodstain in forensic medicine.
