不同生长阶段软骨细胞构建同种异体软骨的研究 被引量:16

Construction of allogeneic cartilage utilizing chondrocytes from difference phase

摘要 目的 研究不同生长阶段山羊软骨细胞 ,形成同种异体组织工程化软骨组织的能力 ,为软骨组织工程寻找种子细胞来源。方法 取胎羊 (孕 10 0d)、新生羊 (10d龄 )及成年羊 (9个月龄 )各大关节软骨组织。酶消化法获取软骨细胞与生物材料Pluronic形成复合物 ,接种于异体羊 (9个月龄 )腹部皮下。按不同时间段取材。所取标本进行大体观察、称重 ,并作组织学检测 ,对新生的软骨组织进行评价。结果 各组接种处均有组织工程化软骨组织形成。胚胎组新生软骨的重量最大 (81.8± 17.9)mg及存留时间最长 (>12个月 )。结论 胚胎软骨细胞可以作为组织工程化软骨的种子细胞来源。 Objective To explore the possibility of the allograft of tissue engineered cartilage in which chondrocytes were isolated from adult,neonatal and fetal goats respectively,and to evaluate the characteristic of the allogeneic tissue engineered cartilage.Methods Chondrocytes were isolated enzymatically from the articular cartilage in fetal (100 days in middle pregnancy),neonatal (10 days) and adult (9 months) Chong Ming goats respectively.Tissue-engineered cartilage was produced using injectable pluronic F-127 polymer mixed with fresh goat articular chondrocytes.The polymer condrocyte mixture in a volume of 0.5 ml containing 5×10 7/ml cells was injected subcutaneously to abdomen in another receipt adult sheep.Samples were harvested at the 2nd,4th,6th,8th week and the 3rd,4th,6th,9th,12th month after transplantation for histologic and transmission electron microscopic detection.Results Histologic evaluation revealed the formation of cartilaginous tissues in the subdermis of abdomen.The allograft of engineered cartilage from fetal chondrocytes have be present in the receipt site for at least 12 months and maintained weight (81.8±17.9) mg.Conclusion Fetal chondrocytes can be provided as seeding cells in cartilage tissue engineering.
