摘要 目的 研究P53、P63、P73蛋白在骨肉瘤中的表达,探讨它们之间的相关性及其在骨肉瘤发生发展中的可能作用。方法 应用免疫组织化学方法和图像分析技术检测52例骨肉瘤标本P53、P63和P73蛋白的表达。结果 骨肉瘤P53、P63、P73蛋白表达水平高于正常骨组织,差别均有显著意义(P<0.01),三者表达与患者性别、年龄及病理类型无关;P53、P63、P73蛋白在骨肉瘤病理Ⅰ级的表达明显低于Ⅱ、Ⅲ级(P<0.05);P73蛋白过表达与肿瘤直径大小及侵犯转移有关(P<0.05),P63与P53蛋白,P73蛋白与P53蛋白表达之间存在着正相关(r=0.328,0.346,P<0.05)。结论 P53、P63、P73蛋白以高表达方式参与骨肉瘤发生发展,并与骨肉瘤进程密切相关。p53基因突变可能与p63、p73基因表达异常有关。 Objective To investigate the expression of p53 , p63 , p73 protein in osteosarcoma, the relationship among these three factors and their possible effects on carcinogenesis and progression of osteosarcoma. Methods Immunohistochemical technique together with computerized image analysis was used to detect the expression of p53 ,p63 , p73 protein in 52 samples of ostersarcoma. Results The expression level of p53 ,p63 and p73 protein in osteosarcoma was higher than that in normal bone tissues (P < 0. 01 ) . There was no correlation between these three proteins and other factors such as age, sex and pathological types. Expression of p53 , p63 and p73 in pathological grade I was significantly lower than those in pathological grade Ⅱ , Ⅲ (P <0. 05) , expression of p73 was correlated to size of tumor and metastasis (P < 0. 05 ) . A significant positive correlation can be identified between p63 and p53 , p73and p53 protein expression ( r =0. 328 , 0. 346 , P <0. 05). Conclusion High expression of P53, P63 and P73 may involve in the carcinogenesis and the development of osteosarcoma and correlates closely to the degree of osterosarcoma. Abnormal expression of P63 P73 may be due to mutation of P53.
