摘要
目的 :建立高效液相法测定野马追中槲皮素的含量。方法 :药材经提取水解后测定其槲皮素的含量来确定野马追中总黄酮的含量 ;采用高效液相法对影响野马追药材中黄酮类化合物提取和水解的影响因素进行了优选 ,;色谱柱 Kro-masil- C18(4.6 mm× 2 5 0 mm ,5μm) ,流动相为乙腈 - 0 .1%三氟乙酸 (2 4∶ 76 ) ,检测波长 36 0 nm,流速 1.0 ml·min- 1。结果 :槲皮素在 5 .6 2 5~ 16 2 mg·L- 1线性关系良好 (r=1.0 0 0 0 ) ,平均回收率为 98.73% ,RSD=1.2 7%。结论 :该方法快速简便 ,重现性好 。
Objective:A RP-HPLC method was established for the determination of quercetin in Eupatorium lindleyanum.Methods:HPLC was used to determine the content of quercetin in Eupatorium lindleyanum after herb was extracted and hydrolyzed. HPLC was used to evaluate the influence of different variables affecting the extraction and hydrolysis of flavoids in herb. A Komasil-C18 (4.6 mm×250 mm, 5 μm) was used mobile phase being acetonitrile-water-TFA (24-76- 0.1 ), flow rate being 1.0 ml·min -1 , detecting wavelength being 360 nm.Results:The calibration curve was linear in the range of 5.625~162 mg·L -1 of quercetin with correlation coefficient being 1.000 0. The average recovery was 98.73%( RSD =1.27%, n =6). Conclusion:The method is quick and simple with good repetition and can be used for quality control.
