摘要 目的 :以99mTc ECD为示踪剂观察神农缬草提取物中挥发油对小鼠脑微循环灌流量的影响。方法 :将 80只小鼠随机分为第一大组和第二大组 ,每大组各 40只。第一大组 40只小鼠随机分为阴性对照组、缬草小剂量组、缬草大剂量组和川芎嗪组 ;阴性对照组以生理盐水灌胃。缬草低剂量组、缬草高剂量组和川芎嗪组分别按 2 0 0、3 0 0和 2 5mg/kg灌胃给药 ,3 0min后经尾静脉注射99mTc ECD示踪剂 (每只 740kBq) 0 .1ml,2 0min后行血、脑放射计数值检测。第二大组 40只小鼠分组及给药方法同第一大组 ,灌胃给药 3 0min后 ,各小鼠按每只 10 μg的剂量腹腔注射去甲肾上腺素 ,5min后尾静脉注射相同剂量的示踪剂 ,2 0min后行血、脑放射计数值检测。结果 :缬草提取物大剂量组小鼠大脑放射计数及脑 /血比值均明显高于阴性对照组 (P <0 .0 5和P <0 .0 1) ;注射去甲肾上腺素后 ,缬草提取物大、小剂量组小鼠大脑放射计数及脑 /血比值亦明显高于脑缺血对照组 (P <0 .0 5和P <0 .0 1)。结论 :神农缬草提取物中挥发油具有显著增加脑组织血流灌注的作用。 Objective:Using 99m Tc ECD as the tracer to evaluate the effects of the extract from valeriana officinalis on rat brain blood flow perfusion.Methods:Eighty rats were divided into group Ⅰ and group Ⅱ.Forty rats of group Ⅰ were randomly divided into negative group for contrast,valeriana officinalis group (low and high dose ) and ligustrazine group.Rats in negative group fed by normal saline,rats in valeriana officinalis group (low,high dose) were fed by extract from valeriana officinalis (200mg/kg and 300mg/kg,respectively ).Rats in ligustrazine group were fed by ligustrazine (25mg/kg). 99m Tc ECD was infused at 30min after the extract feeding.The radioactivity of brain tissue and blood were measured and ID% of brain tissue was calculated.Forty rats of group Ⅱ were divided and feeded as the group Ⅰ,noradrenaline (10μg/meas) was injected at 30min after the extract feeding, 99m Tc ECD was infused at 5min after injection.The group Ⅰ was the same as group Ⅱ of measuring.Results:In the normal rats the ID% of brain tissue and the ratio of brain/blood in treated groups were significantly higher than that in normal controls ( P <0.05 and <0.01 respectively).In the cerebral ischemic rats, similar findings as in normal rats were obtained.Conclusion:The extract from valeriana officinalis may have the effect to increase cerebral blood flow perfusion.
