吸血和约氏疟原虫感染对AdPPOs基因家族转录水平的影响

摘要 目的 观察血餐和约氏疟原虫感染对AdPPO1、AdPPO2和AdPPO3基因转录水平的影响。方法 同批 3~ 5d龄大劣按蚊分为不吸血组 (N)、吸正常血组 (NB)和吸感染血组 (IB) ,在血餐后 2 4h分别制备各组蚊总RNA ,所有样品进行RT PCR后 ,各吸取 10 μlPCR产物进行琼脂糖凝胶电泳 ,凝胶图像分析系统扫描并读取数据 ,并对所得数据进行统计学处理。结果 与不吸血组比较 ,吸血组和感染组AdPPO1的转录水平显著下降 (P <0 0 5 ) ,但吸血组和感染组间没有显著性差异 (P >0 0 5 )。吸血组AdPPO2的转录则明显上调 (P <0 0 5 ) ,但与感染组间没有显著性差异 (P >0 0 5 ) ;AdPPO3基因的转录水平在 3组间均没有显著性差别 (P >0 0 5 )。结论 血餐明显抑制AdPPO1基因的转录 ,但明显上调AdPPO2基因的转录水平 ;约氏疟原虫感染后 2 4h ,AdPPO1。 Objective To observe the effects of blood feeding and Plasmodium yoelii infection on the transcript abundance of AdPPO1, AdPPO2, and AdPPO3 from Anopheles dirus . Methods Anopheles dirus mosquitoes of 3 5 d old were divided into non blood feeding group (N), normal blood feeding group (NB) and Plasmodium yoelii infected blood feeding group (IB). Total RNA from mosquitoes of each group was prepared at 24 h after blood feeding. All samples were analyzed by RT PCR, and the same volume (10 μl) of the PCR products was used for agarose gel electrophoresis. The data obtained were analyzed statistically. Results The transcript abundance of AdPPO1 in groups NB and IB was statistically lower than that in group N ( P <0.05). There was no significant difference between group NB and IB ( P > 0.05). The transcript abundance of AdPPO 2 in group NB was obviously higher than that in group N ( P < 0.05), but no difference was found between groups NB and IB ( P >0.05). There was no significant difference in the transcript abundance of AdPPO3 between the three groups ( P >0.05). Conclusion Blood feeding can inhibit the transcript of AdPPO1, but obviously up regulate the transcript abudance of AdPPO2. However, at 24 h after Plasmodium yoilii infection, blood feeding has no effect on the transcript abundance of AdPPO1, AdPPO2, and AdPPO3.
