间接抗原递呈途径在同种异体表皮细胞移植排斥反应中的作用研究

摘要 目的 探讨间接同种异型识别在体外培养的同种异体表皮细胞移植排斥反应中的作用。方法 体外分离培养人表皮细胞 (Epidermalcells ,EC)、异体人外周血淋巴细胞 (Peripheralbloolymphocytes ,PBL)和单个核细胞 (Peripheralbloodmononuclearcells ,PBM ,含单核细胞 )。将毒性T淋巴细胞相关抗原 4(CTLA4Ig)腺病毒载体转染EC。转染与未转染的EC中均加入异体PBL或PBM共同培养 ,应用氚标记胸腺嘧啶脱氧核苷 ( 3H TdR)掺入法 ,测定各培养系中PBL、PBM增殖情况。结果 CTLA4Ig腺病毒载体能成功转染EC并表达相应蛋白。未转染的EC刺激异体PBL轻度增殖 (P <0 .0 5 ) ,以及刺激含单核细胞的PBM明显增殖 (P <0 .0 1) ;经CTLA4Ig腺病毒载体转染的EC刺激PBL、PBM增殖则明显减弱 (P <0 0 5 )。结论 间接同种异型识别在EC排斥中发挥了重要作用 ,CTLA4Ig能明显减轻该作用。 Objective To explore the effect of the indirect allo recognition on the rejection of epidermal cells. Methods Human epidermal cells(EC), allogeneic human peripheral blood lymphocytes(PBL) and mononuclear cells(PBM, including monocytes) were isolated and cultured in vitro . ECs were transfected with CTLA4Ig adenovirus vector. Allogeneic PBLs or PBMs were added to the transfected and non transfected ECs with simple cultured PBLs or PBMs as the control. The proliferation of PBLs and PBMs was determined by 3H TdR incooperation. Results ECs were successfully transfected with CTLA4Ig adenovirus vector and expressed corresponding proteins. The non transfected ECs could stimulate slight proliferation of allogeneic PBLs( P <0.05) but remarkable proliferation of PBMs(including monocytes) ( P <0.01). The proliferation reaction of PBLs and PBMs decreased significantly( P <0.05) after the stimulation of ECs transfected with CTLA4Ig adenovirus vector. Conclusion Indirect allo recognition might play an important role in the rejection of epidermal cell, but the role can be markedly decreased by CTLA4Ig.
