微波对大鼠脑线粒体细胞色素C氧化酶亚基转录水平的影响 被引量:9

摘要 目的 观察微波辐照对大鼠大脑皮层和海马组织线粒体编码基因COXⅠ和核编码基因COXⅣmRNA表达水平的影响 ,探讨微波辐照是否影响线粒体能量代谢。方法 平均功率密度为 3mW /cm2 和 3 0mW /cm2 微波急性辐照大鼠 1h后 ,应用RT PCR检测大鼠大脑皮层和海马组织COXⅠ和COXⅣ基因转录水平的变化。结果  3mW /cm2 微波辐照后 0、3、2 4h ,大鼠脑海马和大脑皮层COXⅠ、COXⅣmRNA表达无显著变化。 3 0mW /cm2 微波辐照后 0、3、2 4h ,大鼠脑海马和大脑皮层COXⅠmRNA表达均明显降低 ,而COXⅣmRNA表达无显著变化。结论 微波辐照下调大鼠大脑皮层和海马线粒体编码基因COXⅠmRNA表达 ,并存在剂量依赖关系 。 Objective To explore the molecular mechanism of mitochondrial injury induced by microwave irradiation Methods Rats were exposed to microwave irradiation for 1 hour at average power density of 3 mW/cm 2 or 30 mW/cm 2 After microwave irradiation, changes of COXⅠ and COXⅣ mRNA expression level in cerebral cortex and hippocampus in rats were determined by RT PCR Results After 3 mW/cm 2 microwave irradiation for 0, 3 and 24 h, no significant changes of COXⅠ and COXⅣ mRNA expression levels in cerebral cortex and hippocampus in rats were found However, after 30 mW/cm 2 microwave irradiation for 0, 3 and 24 h, COXⅠ mRNA expression in cerebral cortex and hippocampus in rats decreased significantly, but no significant change of COXⅣ mRNA expression levels was found Conclusion Microwave irradiation can down regulate COXⅠ gene encoded by mitochondrial DNA in cerebral cortex and hippocampus in rats in a dose dependent manner
