摘要
目的探讨雌性大鼠心内神经节细胞中ER与ChAT共存的可能性。方法采用免疫组织化学SABC法双重标记技术 ,用鼠抗ER血清和羊抗ChAT血清分别孵育切片 ,葡萄糖氧化酶 -硫酸镍胺或DAB/AP -red试剂盒呈色 ,前者的反应产物呈黑兰色或棕黄色 ,后者的反应产物呈鲜红色。结果在ER/ChAT双重免疫组织化学反应下可见两种细胞 :①ER/ChAT双标细胞 :胞核为ER -IR阳性 ,反应产物棕黄色 ,胞浆为ChAT -IR阳性 ,反应产物鲜红色 ;②ChAT单标细胞 :胞浆为ChAT -IR阳性 ,反应产物鲜红色 ,胞核未见着色 ;在ER和ChAT双重染色的切片上没有发现ER的单标细胞 ;ER -IR阳性神经元约占ChAT -IR神经元总数的 2 7%。结论大鼠心内神经节细胞中ER -IR阳性神经元均为胆碱能神经元。
Objective In order to investigate the coexistence of estrogen receptor(ER) and choline acetyltransferase(ChAT) in intracardiac ganglionic neurons Methods Double labelling immunohistochemical SABC technique was utilized:Separated incubation of the atrial sections were made with mouse anti-ER serum and goat anti-ChAT serum.Results The ER immunoreaction showed dark-blue or brown products in the nuclei.The ChAT-immunoreaction showed pink products in the cytoplasm.There were two kinds of ER/ChAT immunoreactive cells:(1) ChAT-IR single labelled cells,(2) ER/ChAT double labeled cells , accounting for about 27 percent of all the ChAT-IR postive neurons.Conclusion The results reveal that ER-IR neurons are all cholinergic neurons in the intracardiac ganglia in female rats.
