摘要 目的 :观察氧化低密度脂蛋白 (ox LDL)对血管平滑肌细胞 (VSMC)基质金属蛋白酶 9(MMP 9)表达的影响及凝集素样氧化低密度脂蛋白受体 1(Lox 1)在其中的作用。方法 :人脐带动脉平滑肌细胞体外原代培养 ,传至 4~ 5代用于实验。ox LDL干预 ,RT PCR测定VSMCMMP 9RNA和Lox 1mRNA表达。并观察予以Lox 1的抑制剂多聚肌苷酸作用后 ,VSMCMMP 9mRNA的表达的变化。结果 :①ox LDL作用后 ,VSMCMMP 9mRNA表达升高 ,P <0 .0 1,作用 1h出现峰值。ox LDL浓度升高 ,MMP 9mRNA表达增强。②ox LDL作用后 ,VSMCLox 1mRNA表达升高 ,且随着ox LDL浓度的升高有升高趋势 ,均P <0 .0 1。③多聚肌苷酸抑制Lox 1后 ,MMP 9mRNA表达明显下降 ,P <0 .0 1。结论 :ox LDL明显促进VSMCMMP 9表达 ,Lox Objective:To observe how ox-LDL impact MMP-9 m-RNA expression of VSMC and how Lox-1 act as in inflammation course.Method:Smooth muscle cell from human umbilical artery was cultured in vitro. 4~5 th generation were took for use. Grouping according to different ox-LDL concentrations and reaction times. Then, detecting the quantity of VSMC MMP-9 mRNA and Lox-1 mRNA with RT-PCR. Furthermore, observing the change of VSMC-MMP-9 m-RNA expression after giving Lox-1 inhitor-polyinoslnic acid. Result:①The quantity of VSMC MMP-9 mRNA increased highly after co-cultured with ox-LDL, P< 0.01, The peak value was found at about 1th hours. The expression of MMP-9 mRNA was increased as ox-LDL concentration ascending. ② The quantity of VSMC Lox-1 m-RNA increased highly after co-cultured with ox-LDL, P< 0.01. ③ MMP-9 mRNA was reduced when Lox-1 was inhibited by polyinosinic acid, P< 0.01.Conclusion:ox-LDL can induce increased expression of MMP-9 m-RNA of vsmc. Lox-1 plays a vital rule in this process.
