摘要 目的 探讨P2x受体亚型mRNA在大鼠正常和不稳定逼尿肌中的表达变化。方法 建立逼尿肌不稳定大鼠(detrusorinstability ,DI)模型 ,设正常对照 ,以 β actin为内参照 ,RT PCR检测P2X受体各亚型在膀胱中的表达变化。 结果 DI大鼠和正常大鼠逼尿肌中未检测到P2x6 受体mRNA ;P2x1 、P2x5和P2x7受体亚型mRNA表达降低 ;P2x2 、P2x4 表达量显著增加 (P <0 .0 1) ;P2x3未发现明显变化。讨论 DI大鼠逼尿肌内P2x受体部分亚型mRNA表达发生变化 。 s: Objective To study the changes of mRNA expression of P2x receptor subtypes in normal detrusor and detrusor instability (DI) rats. Methods Wistrar rats model of detrusor instability was established by ligating the proximal urethra. Normal control group was employed, and β actin served as the internal control. Changes of the expression of P2x receptor subtypes in the bladder tissue were detected by reverse transcriptive polymerase chain reaction (RT PCR). Different mRNA expressions of P2x receptor subtypes were identified. Results No expression of P2x 6 receptor mRNA was detected in the detrusors of DI and normal rats. Expression levels of P2x 1, P2x 5, and P2x 7 were significantly decreased, but P2x 2 and P2x 4 significantly increased in DI rats ( P <0.01). No change of P2x 3 was detected. Conclusion Changes of mRNA in part of P2x receptor subtypes in DI rats may play an important role in the pathogenic mechanisms of detrusor instability.
