摘要
目的 : 检测人血丙种球蛋白、纯化的抗角蛋白自身抗体 (AKautoAb)、鼠抗人角蛋白 10单克隆抗体 (AK10 )及鼠抗人角蛋白 5和 8单克隆抗体 (AK5&8)对角质形成细胞增殖的影响。方法 : 亲和层析法从丙种球蛋白中纯化了AKautoAb ,四甲基偶氮唑蓝比色法 (MTT法 )检测各处理组无血清培养的角质形成细胞的活性。结果 : 人血丙种球蛋白、纯化的AKautoAb、AK5&8均可抑制角质形成细胞的增殖 ,AK10则无影响。结论 : 人血丙种球蛋中AKautoAb是抑制角质形成细胞增殖的主要成分 。
Objective: To determine the effects of human blood γ-Globulin, purified anti-keratin autoantibody (AK auto Ab ), monoclonal antibody against keratin 10 (AK10) and monoclonal antibody against keratin 5&8 (AK5&8)on proliferation capacity of cultured keratinocytes. Methods: AK auto Ab was purified from human blood γ-Globulin using keratin by Affinity Chromatography. The cultured keratinocytes were treated with human blood γ-Globulin, purified AK auto Ab, AK10 and AK5&8 separately. The proliferation capacity of treated keratinocytes was assessed by MTT determination assay. Results: The proliferation capacity of cultured keratinocytes was inhibited significantly by human blood γ-Globulin purified AK auto Ab and AK5&8. AK10 had no significant inhibitory effect on proliferation capacity of cultured keratinocytes. Conclusion: AK auto Ab plays a major role in the inhibitory effects of human blood γ-Globulin. Monoclonal antibodies against keratin with different molecular weight have different effects on proliferation capacity of keratinocytes.
