摘要
目的 研究一氧化碳与一氧化氮对局灶性缺血脑组织脑含水量及血脑屏障通透性的影响。方法 将 SD大鼠随机分为 4组 :为生理盐水组、Hemin组、Zn PP组及 Hemin+ Zn PP组 ,分别用等量生理盐水、Hemin、Zn PP、Hemin+ Zn PP腹腔注射 ,1h后制成 MCAO模型。栓塞后 2 4h检测血浆 CO及 NO浓度、脑组织含水量、血脑屏障通透性。结果 与生理盐水组相比 ,Hemin组血浆 CO浓度明显升高 ,NO浓度无变化 ,脑组织含水量下降 ,血脑屏障通透性减低 ;Zn PP组血浆 CO浓度明显减低 ,NO浓度上升 ,脑组织含水量上升 ,血脑屏障通透性增高 ;同时注射 Hemin+ Zn PP时 CO浓度无变化 ,NO浓度升高 ,脑组织含水量上升 ,血脑屏障通透性增高。结论 NO及 CO在缺血早期对脑组织均具有保护作用 ,两者之间存在着相互作用 ,CO通过 HO影响 NOS及 NO。
ObjectiveTo evaluate the effect on focal cerebral ischemic tissue in SD rats made by carbon monoxide and nitric oxide. MethodsSD rats were divided into four groups randomly,which including hemin?ZnPP?hemin+ZnPP group and sal ine control group. saline?hemin?ZnPP were injected intra-peritoneal respecti vely 1 hour before middle cerebral artery being occluded(MCAO),24 hours after o cclusion,concentration of CO and NO in blood?brain water?permeability of blood brain barrier(BBB) were examined. ResultsIn Hemin group CO in blood went up,NO in blood had no change,brain water?permeability of BBB redu ced contrast to saline group;in ZnPP group,CO in blood went down,NO in blood goes up,brain water?penetrat ion of BBB rose contrast to saline group;in Hemin+ZnPP group,CO in blood has no ch ange,NO in blood elevated,brain water?permeability of BBB decreased contrast to saline group. Hemin?ZnPP and Hemin+ZnPP had no effect on brain water and permeability of BBB in non-infarcted side( P > 0.05 ). ConclusionBoth carbon monoxide and nitric oxide play a role in brain protection in early focal cerebral ischemia. There is inter action between them. CO may influence NOS and NO by way of HO.
