摘要
静脉注射重组肿瘤坏死因子(rTNF)对小鼠的毒性呈剂量依赖性,多次给药无明显蓄积毒性。rTNF对小鼠S180实体型经静脉、腹腔、皮下、局部给药均有相同抑瘤作用。rTNF体外对肿瘤细胞P388、K562、L1210S180的DNA合成有不同的抑制作用。P388最敏感。体外对细胞的杀伤作用呈浓度—效应反应。无细胞周期特异性。rTNF在体内对巨噬细胞有很强的激活作用,对NK活性影响不明显。
The toxicity and antitumor activity of recombinant tumor necrosis factor (rTNF)appeared to be dependent on doses. The multiple low active doses of rTNF did notinduce accumulated toxicity. The administration of i.v., s.c., 1.p. and in tumor massinjection produced similar antitumor effects. rTNF could strongly inhibited incorpo-ration of ~8HTdR into P388, L1210.K562.S180 cells. Oytotoxic activity of rTNF wasshown dose-effect relationship on P388 cell in vitro. rTNF did not induced changesof cell cycle on P388 cell. rTNF is a powerful agent on macrophage activation in vivo.
