摘要
目的 :观察人白细胞介素 18(IL 18)基因转导对乳腺癌细胞同质粘附能力的影响。方法 :将携带人IL 18基因的质粒pcDNA3.1 hIL 18转染到乳腺癌细胞系Bcap37细胞中 ,通过药物GENETICIN(G 4 18Sulfate)进行筛选 ,利用RT PCR、ELISA法对IL 18的表达水平进行检测。对基因转导后细胞的同质粘附能力进行了测定。结果 :IL 18基因成功的转导入Bcap37细胞中 ,且能持续表达 ;RT PCR电泳结果显示IL 18基因在mRNA水平有表达 ;ELISA结果显示 ,10 6个转导细胞在 2 4h内分泌IL 18的含量是 (187.5± 12 .3)pg ;空载体转染的细胞未检测到IL 18;粘附曲线显示Bcap37 IL 18组的粘附率在各个时间段均明显升高 ,但是空载体组和空白对照组相比之间无明显差异 .。结论 :人IL 18基因成功的整合到人乳腺癌Bcap37细胞基因组中并能持续表达 ;IL
Objective: To study the effect of interleukin 18(IL-18) gene transfer on the adhesion of breast cancer cell Bcap37.Methods:Human breast cancer cell line Bcap37 was transfected with lipofectamine2000 and selected by G418, the expression of rhIL-18 was tested by RT-PCR and ELISA methods, and the changes of adhesion of IL-18 gene-modified cells were detected.Results:The hIL-18 cDNA was successfully integrated into Bcap37 cells, it was 187.5 12.3pg hIL-18 secreted by one million transduced cells in 24 hours; the curve showed that the adhesion of Bcap37-IL-18 cells was enhanced.Conclusion:The IL-18 gene were successfully integrated into Bcap37 cells and can secret hIL-18; the biological characteristics of IL-18 gene-modified Bcap37 cells may be changed.
