摘要 在对实验系统和实验方法的准确性和精密度进行评价的基础上,建立了反相高效液相色谱分析克拉霉素含量的方法.色谱条件:KromasilC18(4.6mm×150mm)反相色谱柱,前接Alltech预柱;甲醇和0.05mol/L磷酸二氢钾(体积比7∶3)的混合液作为流动相,用磷酸调节pH=4.0,流速1.0mL/min;检测波长210nm.该方法简单、快速、灵敏,具有很好的稳定性和精密度. Reversed phase high performance liquid chromatographic method was developed to analyse the content of clarithromycin, and the stability of the method evaluated. The chromatographic conditions in use were as follows: Kromasil C_(18)(4.6 mm×150 mm)column with Alltech guard column; the mobile phase is CH\-3OH and 0.05 mol/L KH_2PO_4(7∶3, V∶V ), pH value adjusted to 4.0 with phosphoric acid, the flow rate is 1.0 mL/min; and the detection wavelength is 210 nm. The results showed that the method adopted was a simple, quick one with fine stability and high accuracy, and can be used to monitor the synthesis of clarithromycin.
