HIV-1整合酶及其抑制剂研究进展 被引量:3

摘要 人免疫缺陷病毒 (Humanimmunodeficiencyvirus- 1 ,HIV 1 )复制周期中的整合过程 ,是将HIV 1DNA整合入宿主DNA的过程 ,也是HIV 1复制周期的不可缺少的过程。HIV 1整合酶 (integrase ,IN)参与整合全过程 ,催化整个整合反应 ,是HIV 1复制过程必不可少的酶 ,也是病毒稳定感染必不可缺的酶。抑制该酶活性将能有效的抗HIV 1。以前 ,由于HIV 1IN溶解等困难、技术理论的滞后 ,限制了我们对此酶的了解。近年来 ,由于分析、分子生物学新技术新方法的应用 ,使它的结构信息、作用机理一步步被揭开。现IN已成为抗HIV 1研究中非常有吸引力的新的靶点。本文阐述了HIV 1整合酶的生化性质包括其结构、作用的分子机理、可能的抑制部位 。
