摘要
目的探讨核因子-κB(NF-κB)、癌基因C—myc和细胞间黏附分子(ICAM-1)的活性表达在大肠癌发生、转移中的作用及其临床意义。方法采用免疫组化S-P法测定50例大肠癌、16例大肠腺瘤、9例大肠腺瘤癌变和8例正常大肠组织中NF-κB p65、C-myc和ICAM-1的表达。结果 大肠癌、大肠腺瘤癌变组织中NF—κB p65、C-myc的表达明显高于大肠腺瘤(P<0.01),C—myc在正常大肠组织中无表达;NF-κB p65、C-myc在大肠癌中的表达呈显著正相关(r=0.35,P<0.01);NF-κB p65和ICAM-1在有转移的大肠癌组织中的表达比无转移的大肠癌显著增加(P<0.01),且两者与大肠癌的转移呈显著正相关(r=0.29,P<0.01)。结论 NF-κB通过对C—myc和ICAM-1的转录调控在大肠癌的发生、转移过程中发挥重要作用,可能将成为大肠癌治疗的一个新靶点。
Objective To explore the expression of the nuclear factor-kB, C-myc and ICAM-1 in human colorectal cancer and the relationship between NF-kB and the occurrence,the metastasis of human colorectal cancer, and to analyze the clinical significance. Methods Fifty patients with colorectal cancer which had been resected by operation, 16 cases of colorectal adenomas,9 cases of carcinomatous change of colorectal adenoma and 8 cases of normal colon were studied.The expression of NF-icB p65, C-myc and ICAM-1 in the tumors and normal colon were examined by immunohistochemical method.Results NF-kB p65,C-myc were expressed more strongly in the colorectal cancer and carcinomatous change of colorectal adenomas than in colorectal adenoma; C-myc was not expressed in normal colon; NF-kB p65 and C-myc had significant positive correlation with colorectal cancer; NF-kB p65, ICAM-1 were expressed more strongly in the colorectal cancer which had metastasis than in that had no metastasis, and they had striking positive correlation with metastasis of colorectal cancer. Conclusion NF-kB plays an important role in the occurrence and metastasis of colorectal cancer through controlling the transcription of C-myc and ICAM-1, and may become a new target for treatment of colorectal cancer.
