摘要
目的 研究碱性成纤维细胞生长因子(bFGF)在胃腺癌组织中的表达,进一步探讨bFGF在胃腺癌血管形成中的作用,为临床正确判断肿瘤细胞的生物学行为及估计患者预后筛选可靠的生物学指标。方法收集2001年1月~2002年2月酒钢医院胃腺癌手术标本蜡块54例。其中低分化胃腺癌32例,高分化22例。采用S-P免疫组化法,严格按照S-P超敏试剂盒的说明操作,显微镜下观察bFGF在胞浆/胞核中的表达。并结合F-ⅧR Ag检测,测量MVD值。结果bFGF在低分化腺癌表达率为75.0%,高分化表达率为36.4%,两者差异有显著性(P<0.05)。bFGF阳性表达组肿瘤MVD高于阴性表达组(P=0.0313)。结论bFGF有助于胃腺癌间质血管形成,且bFGF与癌组织分化程度相关。
Objective To investigate the relationship between tumor angiogenesis and the expression of basic fibroblast growth factor (bFGF) in gastric adenocarcinoma. Methods Expression of bFGF in 54 gastric adenocarcinoma tissue samples,including 32 low differentiated gastric adenocarcinoma cases and 22 well differentiated gastric adenocarcinoma cases, was investigated using immunohistochemical staining.The microvessel density(MVD) was determined by immunostaining for factor VIII-related antigen. Results Of 32 low differentiated gastric adenocarcinoma tissue samples,24(75.0%) were evaluated as bFGF-positive. Of 22 well differentiated gastric adenocarcinoma tissue samples,8(36.4%) were evaluated as bFGF-positive. The expression of bFGF was significantly related with degrees of differentiation of gastric carcinoma( P < 0.05). The microvessel density was significantly higher in bFGF-positive tumours than in bFGF-negative tumors ( P = 0.0313) .Conclusions It is suggested that expression of bFGF plays a role in the promotion of angiogenesis in human gastric adenocarcinoma.
