黄伞多糖的提取及对小鼠腹腔巨噬细胞的激活效应研究 被引量:24

摘要 目的 探讨黄伞多糖的提取方法及对小鼠腹腔巨噬细胞的激活作用。方法 采用水提醇沉的方法提取黄伞多糖 ,体外诱导培养小鼠腹腔巨噬细胞。用ELISA法检测培养上清中的细胞因子 ,Griess试剂检测其NO的含量 ;FACS检测巨噬细胞吞噬功能 ;MTT间接法观察巨噬细胞体外杀伤活性的变化。结果 黄伞多糖组巨噬细胞产生IL 1β及NO的水平明显高于PBS对照组 ,黄伞多糖组巨噬细胞吞噬功能、杀伤活性明显高于PBS对照组 ,两组间各项指标比较均有显著性差异 (均 P <0 0 5 )。结论 黄伞多糖能激活小鼠巨噬细胞 ,可增强巨噬细胞IL 1β及NO产生的水平 ,增强其吞噬功能及体外杀伤活性。 Objective: To explore the dominant activation effect of Pholiota adipdsa(Fr)ouel polysacckarose on peritoneal macrophages in rats. Methods: Cultured Pholiota adipdsa(Fr)ouel amylose was extracted and peritoneal macrophages in rats were induced and reared in vitro. By the method of ELISA, cytolysis was detected in the cultured supernatant, the NO content by Griess reagent, and phagocytosis by FACS. The change in deactivation of the macrophages in vitro was observed by MTT indirectly. Results: IL-1β and NO content produced by the macrophages were significantly higher in the Pholiota adipdsa(Fr)ouel amylose group than those in the control group. The phagocytosis and deactivation of the macrophages were significantly higher than those in the PBS control group. The indexes showed significant difference beteen the two groups (P<0.05). Conclusion: Pholiota adipdsa(Fr)ouel amylose can activate macrophages in rats, enhance the production of IL-1β and NO by the macrophages, and improve their phagocytosis and deactivation as well.
