大鼠骨髓基质细胞传代培养中相关基因的差异表达 被引量:2

DIFFERENTIAL EXPRESSION OF CORRELATIVE GENES DURING SUBCULTURE OF RAT BONE MARROW STROMAL CELLS

摘要 目的 :研究大鼠骨髓基质细胞 (BMSCs)体外传代培养中相关基因的差异表达。方法 :取大鼠骨髓 ,分离、培养骨髓基质细胞并传代 ,抽取细胞 (mRNA)并逆转录成cDNA探针 ,用含有 1 1 76个基因片段的cDNA芯片检测相关基因的差异表达 ,整理、分析实验数据。结果 :传至P3代细胞与原代细胞 (P0 )相比有1 0 4个基因的差异表达超过了 3倍 ,其中有 5 3个基因表达量明显上升 ,5 1个基因表达量明显下降 ,主要涉及一些细胞粘附分子及胞外基质、细胞信号与传导、细胞受体和细胞骨架等方面的相关基因。结论 :通过基因芯片技术可以显示BMSCs在体外传代培养中众多基因的差异表达 ,而这些基因的差异表达与细胞的增殖和分化存在一定的关系 。 Objective:To explore the differential expression of correlative genes during subculture of rat bone marrow stromal cells in vitro . Methods: rBMSCs were acquired from rat bone marrow and cultured with DMEM. cDNA was reverse transcribed from mRNA of the samples, and hybridized with 1176 genes on the microarray membrane. The experimental data were then processed and analyzed. Results: There were 104 genes expressed differentially 3 times in passage 3 cells more than in primary cells, the expression level of 53 genes was up regulated while that of 51 genes were down regulated significantly in P3 cells, with some gene expressions relating to cell adhesion molecule, extracellular matrix, cell signal molecules , cytoskeleton,etc. Conclusion: The cDNA microarray can be used to show the multi gene expression that relates to the cell proliferation and differentiation during subculture of rBMSCs, and provides important bases on the theoretic studies of the stability of BMSCs in subculture and clinical application to the genic level.
