摘要
以结缕草愈伤组织为试材 ,利用农杆菌介导法辅以辐照、愈伤组织状态、共培养条件及负压处理等处理方法对愈伤组织转化频率的影响进行研究。结果表明 :2Gy的辐照剂量利于提高转化频率 ;1- 2个月的愈伤组织适于转化 ;全光照的共培养条件下转化频率高于 16 /8h光照培养和暗培养条件下转化频率 ;农杆菌与愈伤组织混合培养时给予适当的负压条件可提高愈伤组织转化频率和存活率。
The assistant method of agrobacterium-mediated transformable efficiency which includes irradiation, callus, co-cultivation and negative pressure, is explored with the callus of Japanese Lawngrass(Zoysia.japonica steud.). The result is as following: 2 Gy of irradiation-dosage benefits the exaltation of transformable efficiency; 1-2 months of callus proper is fit for the transformation. The transformable efficiency of the whole light-shine to co-cultivation is higher than that of 16/8 h photoperiod and of the whole dark. The proper negative pressure, when agrobacterium and callus cultivated, could increase transformable frequency and survival rate of callus.
