摘要
基因表达文库构建和筛选技术是 80年代初发展起来的用于筛选和克隆基因的基本手段。表达文库中含有大量基因表达信息 ,使其成为广大实验室优先建立的文库之一。迄今为止 ,表达文库的构建方法在传统方法的基础上不断被改善 ,针对各种类型的目的基因克隆的要求 ,可以选择不同的载体和方法来构建适当的基因表达文库。本文详细介绍了多种构建基因表达文库的方法 ,比较它们的构建方法的优点和不足之处 ,并对它们在实际应用中的作用作了较为全面的分析。
Gene expression library is a new advance in screening and cloning novel genes which is developed from early 80's. Since it contains large amount of gene expression information, it becomes the first choice for screening novel genes in most laboratories. In present, based on the traditional expression library construction method, different methods can be used for different purpose of gene cloning. In this paper, many construction methods are introduced and their merits and shortcoming are also compared. Moreover, the application of gene expression library in practice has been introduced as well.
