吲哚美辛对胃癌SGC 7901细胞增殖及Cyclin D1蛋白表达的影响 被引量:1

摘要 目的:观察吲哚美辛对胃癌SGC 7901细胞增殖及细胞周期调控蛋白Cyclin D1表达的影响,探讨吲哚美辛抑制细胞增殖的机制。方法:采用MTT法观察吲哚美辛对胃癌细胞SGC 7901增殖的影响,采用流式细胞仪观察细胞周期分布的变化,采用免疫细胞化学方法检测Cyclin D1蛋白的表达。结果:吲哚美辛呈时间、浓度依赖方式抑制胃癌SGC 7901细胞增殖,改变细胞周期的分布,增加G0/G1期细胞的比例,下调Cyclin D1蛋白的表达。结论:吲哚美辛可能通过下调Cyclin D1蛋白表达,影响细胞周期的分布来抑制胃癌SGC7901细胞增殖。 Objective: We investigate the effects of indomethacin on the cells proliferation and the expression of cell cycle Cyclin D1 protein in human gastric cancer cell line SGC 7901. Methods: after treated with indomethacin, SGC 7901 cell growth was quantified by MTT assay and the expression of Cyclin Dl protein in the cells was detected by immunocytochemical stainning . Flow cytometry was used to observe the influence on the distribution of cell cycle. Results: Indomethacin inhibited the cells proliferation in a time and dose-dependent fashion, altered the distribution of cell cycle and increased the proportion of cells in the G0/G1 phase and down-regulated the expression of Cyclin D1 protein. Conclusion: Indomethacin may inhibit the proliferation of gastric cancer cells SGC7901 through down-regulating the expression of Cyclin D1 and affecting the distribution of cell cycle.
