摘要
目的 探讨转化生长因子 β(TGF β)及其受体在增生性瘢痕形成中的作用机制。 方法 收集临床手术切除后的正常皮肤组织 7例和增生性瘢痕组织标本 11例 ,用免疫组织化学和原位杂交的方法检测TGF β及其TGF β受体 (TGFR)Ⅰ、TGFRⅡ在组织中的定位分布、蛋白和mRNA表达。 结果 在正常皮肤组织中 ,与TGF β1 、TGF β2 、TGFRⅠ比较 ,TGF β3和TGFRⅡ呈较高表达 ;在增生性瘢痕组织中则呈现与正常皮肤组织中的表达相反的结果。与正常皮肤组织相比 ,增生性瘢痕组织中TGF β1 、TGF β2 和TGFRⅠ的蛋白和mRNA表达均明显增加 ,而TGF β3和TGFRⅡ的蛋白以及mRNA表达相对减少。 结论 创面愈合过程中TGF β及其受体不同水平的表达与增生性瘢痕的形成直接相关。
Objective To explore the role of transforming growth factor beta (TGFβ) and their receptors (TGFβ-R) in the pathogenesis of hypertrophic scar. Methods Specimens of normal skin and hypertrophic scar were harvested and the distribution and the expression of the TGFβ and TGFβ-R were determined by immunohistochemistry and in situ hybridization method. Results The expressions of TGFβ and TGFβ-RⅡ in normal skin were higher than the expression of TGFβ1, TGFβ2 and TGF-RⅠ. But in hypertrophic scar the results were on the contrary. The mRNA expressions of TGFβ1, TGFβ2 and TGFRⅠ were evidently increased with decreased mRNA expression of TGF-β3 and TGFR Ⅱ in the hypertrophic scar when compared with those in the normal skin. Conclusion The expression of TGF-β (β1, β2, β3) and their receptors in different levels during the process of wound healing might be related to the formation of hypertrophic scars.
