米非司酮对早孕猕猴蜕膜和绒毛组织绒毛膜促性腺激素-β和Ⅳ型胶原的影响 被引量:2

摘要 目的 :分析米非司酮对早孕猕猴蜕膜及绒毛组织中绒毛膜促性腺激素 β(β CG)和Ⅳ型胶原表达的影响。方法 :与健康雄猴自然交配的、处于月经周期第 38~ 4 5d的经B超确诊受孕的雌性猕猴 18只 ,随机分为米非司酮组 [米非司酮 ,15mg/ (ml·kg) ],空白对照组 (不做任何处理 ) ,赋型剂对照组 (0 .5 %羧甲基纤维素钠 ,1ml/kg ,1次 /d ,连续 3d)。采用免疫组化二步法研究早孕猕猴蜕膜和绒毛组织中 β CG和Ⅳ型胶原的表达。  结果 :与对照组比较 ,米非司酮组早孕蜕膜组织中基质、腺上皮、蜕膜细胞胞质内 β CG免疫反应强度均明显减弱 ,绒毛组织中合体滋养层细胞胞质内 β CG免疫反应强度也明显减弱 ;Ⅳ型胶原在蜕膜组织的腔上皮、腺上皮基膜、蜕膜细胞周围、血管周围、绒毛组织中合体滋养层细胞周围免疫反应强度均明显减弱。 结论 :米非司酮具有强效促蜕膜组织 β CG生物活性下降、绒毛组织β CG合成下降和促绒毛、蜕膜组织Ⅳ型胶原降解的作用。 Objective: To study the effect of mifepristone on the expression of chorionic gonadotropin beta subunit(β-CG)and collagen type Ⅳ in female Rhesus monkey decidua and villus during the first trimester of pregnancy. Methods: Eighteen sexually mature female Rhesus monkeys were allowed to cohabitate with their male counterparts and diagnosed as being pregnant by B-ultrasound at days 38~45 of the menstrual cycle. They were randomly divided into mifepristone treatment group (15 mg/ml·kg·day,3 days), control group(no treatment) and CMC-Na treatment group( 0.5%CMC-Na,1 ml/kg·day, 3days).The content and distribution of chorionic gonadotropin beta subunit and collagen type Ⅳ in the female Rhesus monkeys were examined by immunohistochemical technique. Results: Compared with the control group, the immunohistochemical reaction intensity of chorionic gonadotropin beta subunit in the plasm of stroma cells, glandular epithelial cells and decidual cells of decidua, and synctial trophoblast cells of villus significantly decreased in the mifepristone treatment group. The positive staining of collagen type Ⅳ surrounding the epithelial glandular basal membrane, decidual cells and blood vessels in decidua and surrounding the synctial trophoblast cells in villus were significantly reduced. Conclusion: Mifepristone may have a strong effect in decreasing the bioactivity of β-CG in decidua and the synthesis of β-CG in villus,and in accelerating the degradation of collagen type Ⅳ in decidua and villus.
