Preliminary Study in Flow Cytometric Detecting Anti-platelet Antibody in Whole Blood

摘要 目的 初步建立一种以全血为标本的流式细胞仪 (FCM)检测血小板抗体的方法。方法 FCM多参数分析全血中的CD6 1标记的血小板群。结果 每次检测需血以微升为数量级单位 ,所用时间 2h。能从正常的血小板中检出 3%含有CD6 1抗体的血小板 ,阳性率与理论值呈明显相关 (r >0 .9,P <0 .0 0 1) ;可检测血小板计数低达 10× 10 9/L的标本。结论 本法具有结果准确、重复性好的优点 ,需血量少 ,病人易接受 ,并且更接近生理状态 ;多参数检测 ,快速省时 ,更适用于血小板极低或抗体量少的患者。 Purpose: To develop a method detecting platelet associated immunoglobulin (PAIg) and platelet specific immunoglobulin in whole blood using flow cytometry(FCM), and study the methodological factors affecting flow cytometric analysis of anti-platelet antibody. Compare FCM with traditional means. Methods: Multiparameter FCM analyze platelet in whole blood. Results: The results of sample detected by different checker had no significant difference(P > 0.05). FCM was accurate and had sufficient sensitivity even platelet count was 10 × 109/L. As much as 3% CD61-blocked platelets among normal platelets could be checked out. Conclusions: FCM was accurate and had sufficient sensitivity even platelet count was minim. FCM was able to check out sparklet antibody.
