摘要
目的 探讨生存素 (Survivin)和天冬氨酸特异性半胱氨酸蛋白酶 3 (Caspase 3 )在原发性肝细胞癌 (HCC)组织中的表达及两者在肝癌细胞凋亡中的作用和关系。方法 采用蛋白印迹和免疫组织化学法检测 43例肝癌组织、2 0例癌旁组织和 11例正常肝组织中Survivin和Caspase 3蛋白表达情况。逆转录 聚合酶链反应 (RT PCR)检测Survivin和Caspase 3表达情况。 结果 肝癌组织中Survivin表达率为 69.8% (3 0 /4 3 ) ,而除 1例癌旁组织外 ,其他癌旁组织和正常肝组织中未见表达 ;Caspase 3在癌组织、癌旁组织和正常肝组织中表达率分别为 2 9.4% (18/4 3 )、75 .0 % (15 /2 0 )和 81.8% (9/11)。结论 Survivin在肝癌组织中高表达 ,并通过抑制Caspase 3而抑制肝癌细胞凋亡 ,两者可能是肝癌细胞凋亡信号传导网络中的重要一环。
Objective To investgate the expression ofsurvivin and aspartate-specific cysteine proteases 3 (Caspase-3) and their correlations with the apoptosis in primary hepatocellular carcinoma (HCC).Methods The survivin and Caspase-3 protein expression was evaluated using Western blot and immunohistochemistry.The expression of survivin and Caspase-3 mRNA was detected by using reverse transcription-polymerase chain reaction in 43 HCC tissues,20 para-carcinoma tissues and 11 normal liver tissues.Results Survivin protein was detected in 30 of 43 HCC tissues (69.8 %) and 1 of 20 para-carcinoma tissues (5 %) and none of 11 normal liver tissues; Caspase-3 protein was detected in 18 of 43 HCC tissues (29.4 %),15 of 20 para-carcinoma tissues (75 %) and 9 of 11 normal liver tissues (81.8 %).Conclusion Survivin inhibited apoptosis of HCC cells through suppressing Caspase-3 and thetwo genes were probably an important link in the signal pathway of HCC cells apoptosis.
