摘要
为更好地保护和开发利用黄鳝资源,从40个随机引物中选取30个重复性好、扩增稳定的随机引物,应用RAPD技术对黄鳝属中产自缅甸的山黄鳝、印度尼西亚的穴黄鳝、中国黄鳝的DNA进行遗传多样性分析.共检测到315个位点,3种黄鳝种群间的多态位点比例为85.08%,遗传多样性指数为0.157 5.DPS2003种群遗传分析软件包分析显示,3种黄鳝种群间的遗传距离为0.710 3, 相似度系数为0.289 7,表明3种黄鳝都具有较高的遗传变异.
The samples of three species of synbranchi form eels,Monopterus cuchia,M. fossorius and M. albus were collected from Yangon of Myanmar,Indonesia and YuanJiang County,Hunan Province,China respectively.Randomly amplified polymorphic DNA (RAPD) technique is usded to analyse the specific genetic markers and their genefic diversity of the wild grouper population of four individuals from each species.40 primers of OPA,OPB and OPD were screened,30 primers produced reproducible fingerprints.The percentage of polymorphic loci(P)of interpopulation of three species is 85.08%,with a genetic diversity index (H) of 0.157 5.Based on the RAPD data,the genetic distances among these three species of Monopterus,were computed by using DPS2003 software package.The genetic distance (D) is 0.710 3 and the genetics imilarity index(S) is 0.289 7 among interpopulation three species of Monopterus.The result indicates that the genetic diversity is rich among interpopulation and intrapopulation of three species.The genetic diversity of intrapopulation is higher than that of interpopulation.
