葛根汤和桂枝汤调节椎间盘组织Fas、bcl-2蛋白表达的实验研究

Expression of the Fas receptor,bcl-2 in the cervical intervertebral disc of the rabbit modified by the Guizhitang and Gegentang

目的 :比较检测风寒湿痹证型颈椎病动物模型中颈椎间盘Fas、bcl 2蛋白的表达 ;检测葛根汤、桂枝汤对风寒湿型颈椎病家兔颈椎间盘组织中Fas、bcl 2蛋白的调节作用。方法 :8月龄雄性新西兰白兔 2 4只 ,随机分为正常对照组、风寒湿刺激组、葛根汤治疗组、桂枝汤治疗组。风寒湿刺激组温度 6℃ ,湿度 95 % ,风力 6级 ,每日连续刺激 4h ,间断重复刺激 12 8h ;葛根汤、桂枝汤治疗组在风寒湿刺激组造模基础上服用。采用免疫组织化学ABC法检测椎间盘石蜡切片中Fas、bcl 2蛋白的表达情况。结果 :风寒湿刺激组同正常对照组比较 ,Fas表达均上调 (P <0 0 1) ,葛根汤降低Fas表达 ,与风寒湿刺激组比较有显著性差异 (P <0 0 1)。风寒湿刺激组同正常对照组比较 ,bcl 2表达均下降 (P <0 0 5～ 0 0 1) ,葛根汤上调bcl 2表达 ,但与风寒湿刺激组比较没有显著性差异 (P >0 0 5 ) ,桂枝汤对bcl 2的调节作用不明显。结论 :风寒湿刺激组同正常组对照比较 ,Fas表达均上调 ,bcl 2表达均下降 ,葛根汤降低Fas表达 ,上调bcl 2表达 ,发挥延缓椎间盘退变的作用 ,桂枝汤调节上述细胞因子的作用低于葛根汤。

WT5”BZ][HT5”H]Objective:[HT5”SS]To compare the expression of the Fas receptor,bcl-2 in the degenerative intervertebral disc of the rabbits cervical spondylosis model induced by Fenghanshi.To study the effects of the Guizhitang,Gegentang on the expression of the Fas receptor,bcl-2.[HT5”H]Methods:[HT5”SS]24 male rabbits were divided randomly into normal group,Fenghanshi group,Gegentang group and Guizhitang group.Fenghanshi model was created in 6 ℃ temperature,95% humidity,and 6 grade wind with 4 hours stimulation at each day for the total of 128 hours.Fenghanshi group was treated by Gegentang and Guizhitang.The expression of the Fas receptor,bcl-2 in the degenerative disc was detected with the immunohistochemistry ABC.[HT5”H]Results:[HT5”SS]The expression of the Fas receptor increase in the Fenghanshi model compare with the normal (P<0 01).Gegentang could regulate down the expression of Fas receptor in the fenghanshi model (P<0 01).On the other hand,the expression of the bcl-2 decrease in the Fenghanshi model compared with the normal (P<0 05～0 01).Gegentang could regulate up the expression of Fas receptor in the Fenghanshi model (P>0 01) [HT5”H]Conclusion:[HT5”SS]The expression of the Fas receptor can be regulated up and the bcl-2 regulated down in the Fenghanshi model compared with the normal.Gegentang can regulate down the expression of the Fas receptor and regulate up the bcl-2.[WT5”HZ]
