乙型肝炎患者白细胞介素-2产生障碍的机制探讨

摘要 用白细胞介素2(IL-2)依赖细胞株(CTLL)检测了28例慢性迁延性肝炎患者外周血单个核细胞(PBMCs)产生IL-2的活性;同时用Southern印迹分子杂交技术检测PBMCs内HBVDNA。结果显示患者IL-2活性显著降低(P<0.01)。而HBV DNA检出组又比未检组低(P<0.05)。表明患者IL-2活性降低是多种原因造成的,而PBMCs内HBV DNA的存在可以影响IL-2活性,从而促进HBV感染的慢性化形成。本研究提示,对乙型肝炎患者进行重组IL-2治疗有可能阻止慢性肝炎的进展。 Peripheral blood mononuclear cells (PBMCs) were isolated from 28 Patients with chronic persistent hepatitis B and 10 normal individuals for detecting HBV DNA by Southern blot analysis and assaying interleukin-2 (IL-2) with cytotoxic molecular hybridization T-lymphocyte line (CTLL) which depends on IL-2. IL-2 activities of Patients were markedly decreased in comparision with those of controls (P<0.01). IL-2 activity of the group with detected HBV DNA was significantly lower than that of the group without detected HBV DNA (P<0.05). The results indicated that the existence of HBV DNA in PBMCs is one of the decrease in IL-2 activity, and may play an important role in the immunopathogenesis of such diseases as cause the chronical process. It might be necessary to treat patients with chronic HBV infection by using IL-2.
