摘要
〔目的〕建立环境水样Cu(Ⅱ )的分析新方法。〔方法〕人血清白蛋白荧光猝灭法。〔结果〕根据Cu(Ⅱ )在pH=7.43的KH2 PO4 -Na2 HPO4 缓冲溶液中对HSA荧光的猝灭效应 ,建立了HSA荧光猝灭法测定Cu(Ⅱ )的分析新方法。在实验最适条件下Cu(Ⅱ )浓度在 0 .13 -6.0 0 μg/ml范围内有良好的线性关系 (r =0 .997)。方法检出限 3 8ng/ml;相对标准偏差 6.4%-1.5 %;样品加标回收率 94.5 %-10 1.0 %。〔结论〕方法简便 ,特异 ,灵敏度高。可直接用于环境水样中Cu(Ⅱ )的测定。
Objective To establish a practical method for determination of Cu(Ⅱ) in samples of environmental water.〔Methods〕 Fluorescent quenching method using human serum albumin (HSA) was applied.〔Results〕 The method based on the fluorescent quenching effect of Cu(Ⅱ) to HAS in KH 2PO 4-Na 2HPO 4 buffer solution at pH=7.43.Under the selected condition,the quenching value of I 350 nm was found linear to the concentration of Cu(Ⅱ) in the range of 0.13-6.00μg/ml ( r =0.997).The detection limit was 38ng/ml.The RSD was 0.4%-1.5% and the recovery rate 94.5%-101.0%.〔Conclusions〕The method is simple,acCu(Ⅱ)rate,sensitive and highly selective for determination of Cu(Ⅱ) in samples of environmental water.
