摘要
以紫萁孢子叶小段为接种材料,研究了影响孢子叶小段萌发的培养基组成及影响紫萁原叶体增殖的各项因素、孢子体苗分化的条件和孢子体试管苗的炼苗技术,结果表明:在温度25℃、光照12h·d-1、光照强度2000lx、培养基pH5.8等条件下,较适合原叶体增殖的培养基是1/2MS+KT5.0mg·L-1+IBA1.0mg·L-1+KH2PO4200mg·L-1+活性炭0.5%。其分化出的孢子体苗的原叶体具有质地疏松、片状体大且肉质厚等性状。在温度20~28℃、湿度90%~100%的荫棚或林下,选择基质椰茸进行炼苗,根系粗壮3条以上、叶3片、叶长3cm以上的幼苗成活率可高达97.6%。
Using a small part of Osmunda japonic Thunb spore leaf as inoculated material?熏 we made a research on the composition of culture base which may affect the spore's shooting?熏 all kinds of factors affecting the reproduction of the prothallium?熏 the condition to break up the sporpphyte seedling and the building up seedling techniques of sporpphyte test tube seedling. It shows that with the temperature of 25℃?熏the sunshine 12h·d-1, the sunshine intensity 2 0001x, and with the condition of pH5.8 to the culture base?熏 the better culture base suitable for the propagation of prothallium is 1/2MS+KT5.0mgL·-1 +IBA1.0mg·L-1+ KH2PO4200 mg·L-1+active carbon 0.5%. The prothallium fit to break up the sporpphyte seedling has the feature of loose texture?熏 slice shape and thick meat quality. Under the condition of the temperature 20~28℃?熏humidity 90%~100%?熏 an awning or a forest with four-tenths shadow and six-tenths sunshine ?熏you may choose matrix shredded coconut stuffing to build up seedlings?熏 the seedlings which have more than 3 thick roots?熏 3 pieces of leaves and the length of leaves up to over 3cm can have 97.6% survival rate.
