摘要 目的 :探讨同型半胱氨酸 ( Hcy)对大鼠血管平滑肌细胞 ( VSMCs)核转录因子 κB( NF- κB)活性的影响。方法 :在大鼠主动脉平滑肌细胞培养基中加入终浓度为 0 .2 5 mmol/L的 Hcy,于 30 min、1h、2 h收集细胞 ,提取胞核蛋白。电泳迁移率改变法检测 NF- κB活性的变化。结果 :静息状态下 VSMCs具有一定的 NF- κB活性水平。经Hcy刺激后 ,NF- κB活性迅速增加。密度扫描仪结果提示 ,NF- κB活性 30 min时为对照 1.76倍 ,1h时为对照 1.91倍 ,2 h时为对照 1.84倍 ( P<0 .0 1)。结论 :Hcy能使 VSMCs NF- κB活性增加 ,提示 Hcy对 VSMCs的作用部分是通过 NF- Objective: To investigate the effect of homocysteine (Hcy) on nuclear factor-κB (NF-κB) activity in cultured rat vascular smooth muscle cells (VSMCs). Methods: Rat VSMCs were stimulated with 0 25 mmol/L Hcy. Cells were collected and nuclear protein was extracted at 30 min, 1 h and 2 h following stimulation. NF-κB activity was examined by electrophoretic mobility shift assay (EMSA). Results: Baseline NF-κB nuclear binding in VSMCs increased more than 1 76-fold at 30 min ( P <0.01), 1 91-fold at 1h ( P <0.01) and 1 84-fold at 2 h ( P <0 01) following stimulation of Hcy. Conclusion: Hcy can enhance transient mobilization of NF-κB in VSMCs, which suggests that some regulating effect of Hcy on VSMCs might be exerted through NF-κB activation pathway.
