摘要
目的 了解血管内皮生长因子(VEGF)在新生鼠缺氧缺血性脑损伤后表达的动态变化。方法 采用Rice法制备缺氧缺血性脑病模型,将80只7日龄SD大鼠随机分为:正常对照组、模型后即刻组、1h组、3h组、6h组、12h组、24h组、48h组、72h组、7d组。用免疫组织化学方法观察VEGF表达的动态变化。结果 正常对照组中大鼠脑内几乎无VEGF表达;模型组结扎侧大脑皮质,纹状体,海马在模型后即刻即有VEGF表达,3~12h达高峰,随后开始下降,7d后呈低水平稳定表达。结论缺氧缺血作为一种信号刺激VEGF在神经细胞内表达增加;VEGF的表达增加具有时间依赖性;VEGF对缺氧缺血性神经元损伤可能具有一定的保护作用。
Objective To investigate the time course of changes of Vascular endothelial growth factor(VEGF) expression in newborn rats following hypoxia-ischemic brain damage (HIBD). Methods The hypoxiaischemic brain damage model were made by Rice legal system. The HIBD models were established with seven days old SD rats and all the rats were divided into ten groups at random: contral groups, immediately after hypoxia-ischemic(HI), 1h group, 3h group,6h group, 12h group, 24h group, 7d group. The expression of VEGF was examined by means of immunohistochemistry method. Result VEGF immunoreactive positive cells were not found in the brain tissue of normal group. Expressions were rapidly induced in the cerebral cortex, striatum and hippocampus in the ligate lateral hemisphere brain, and peaked at 3h, remained to 12h, then decreased and stabilized at low level after 7d. Conclusions Hypoxia-ischemic as a signal stimulate the expression of VEGF in nerve cells. The increased VEGF levels are related with time. The expression of VEGF may be one of neuronal protection mechenisms after ischemic cerebral injury.
