利用冰冻切片技术检测大鼠全脑缺血后海马CA1区线粒体细胞色素C变化(英文) 被引量:1

摘要 目的利用冰冻切片技术检测大鼠全脑缺血后海马CA1区神经元线粒体CytC的变化。方法采用4-VO法制作全脑缺血模型,将SD大鼠随机分为假手术对照组、全脑缺血组。观察缺血后24 h海马CA1区细胞色素C(Cytochrome C,CytC)变化。结果缺血后24 h,假手术组海马CA1区CytC呈阳性表达,并且呈现出特异性的点状分布,符合CytC线粒体分布的特征,全脑缺血组海马CA1区CytC表达明显降低。结论利用冰冻切片技术能方便、直观的检测脑缺血再灌注后神经元线粒体CytC的变化,再灌注24小时海马CA1区神经元线粒体CytC明显下降。 Objective: Using frozen section technique to detect the changes of cytochrome c in hippocampal CA1 subregion neurons 24 hours after transient global cerebral ischemia. Methods: Using 4-VO technique, the transient global cerebral ischemia model was made. Sprague Dawley rats were randomed into sham operation group and cerebral ischemia group. Changes of cytochrome c in hippocampal CA1 subregion neurons 24 hours after transient global cerebral ischemia were observed. Results: There was expression of cytochrome c in hippocamp CA1 subregion in sham operation group presenting punctual distribution, 24 hours after transient global cerebral ischemia the expression of hippocampal CA1 subregion in cerebral ischemia group obviously decreased (P<0.01). Conclusions: Frozen section technique can conveniently and visualized detect the changes of cytochrome c of neurons 24 hours after transient global cerebral ischemia. Cytochrome c in hippocampal CA1 subregion neurons 24 hours after transient global cerebral ischemia obviously decreased.
