化疗+G-CSF方法动员外周血干细胞时骨髓及外周血相关细胞亚群的动态变化(英文) 被引量:1

摘要 目的 探讨应用化疗 +G -CSF方法动员外周血干细胞时 ,恶性血液病患者的骨髓及外周血CD3 4 +细胞和T细胞亚群的变化特点。方法  16例拟行自体外周血干细胞移植患者 ,在应用化疗 +G -CSF方法动员外周血干细胞期间 ,应用流式细胞仪测定骨髓及外周血CD3 4 + 和T细胞亚群动态变化情况。结果  1.恶性血液病患者在应用G -CSF前 ,骨髓中CD3 4 + 细胞的初始值为 (0 .5 2± 0 .31) %。应用G -CSF 4 8h后 ,CD3 4 + 细胞测定值为 (1.2 2± 0 .4 2 ) % ,较前有明显增加 (P <0 .0 0 1)。CD3 4 + 细胞的高峰值出现在应用G -CSF 96h后 ,为 (2 .2 3± 0 .34) % ,较未应用G -CSF时差异显著 (P <0 .0 0 1)。 2 .恶性血液病患者在应用G -CSF前 ,外周血CD3 4 + 细胞测定值为 (0 .39± 0 .2 7) %。在应用G -CSF 72h后外周血中CD3 4 + 细胞的测定值为 (1.2 9± 0 .6 4 ) % ,较前有明显增加 (P <0 .0 0 1)。应用G -CSF96h后 ,CD3 4 + 细胞的测定值达高峰 ,为 (1.4 1± 0 .73) % ,较未应用G -CSF前差异显著 (P <0 .0 0 1)。 3.无论是否应用G -CSF ,恶性血液病患者的T淋巴细胞亚群比例均处于倒置状态 ,应用G -CSF后随CD3 4 + 细胞的逐渐增加 ,T淋巴细胞亚群变化不明显 (P >0 .0 5 )。结论 恶性血液病患者 ,在应用化疗 +G -CSF? Objective: To approach what changes happen about CD 34 + cells and T lymphocyte subsets after using chemotherapy plus G-CSF with patients. Methods: Flow cytometry was used to detect the changes of the percentages of CD 34 + cells and T lymphocyte cell subsets in 16 cases of patients when they were treated with chemotherapy and injected with G-CSF for mobilizing peripheral blood stem cells. Results:1. Without using G-CSF, the percentages of CD 34 + cells in the bone marrow of the patients ranged in (0.52±0.31)%. After Using G-CSF for 48 hrs, the percentages ranged in (1.22±0.42)%, obviously increased compared with those of no using G-CSF ( P < 0.001 ). After 96 hrs the highest percentages were reached and ranged in (2.23±0.34)%, significant increased compared with those of no using G-CSF ( P <0.001). 2. Without using G-CSF, the percentages of CD 34 + cells in peripheral blood of the patients ranged in (0.39±0.27)%. After using G-CSF for 72 hours, they ranged in (1.29±0.64)%, obviously increased compared with those of no using G-CSF ( P <0.001). After 96 hrs the highest percentages were reached and ranged in (1.41±0.73)%, significant increased compared with those of no using G-CSF ( P <0.001).3.CD 4/CD 8 of the patients were abnormal (<1) whether in bone marrow cells or peripheral blood cells. There were no obvious changes of T lymphocyte subsets in the patients except that CD 34 + cells increased gradually( P >0.05) when using G-CSF for some days . Conclusions:The dynamic change of the percentages of CD 34 + cells and T lymphocyte subsets in bone marrow or peripheral blood of patients with CR complete remission has itself characteristics after using chemotherapy plus G-CSF for days.
