摘要
目的探讨PDGF与IGF-Ⅰ对体外培养成骨细胞增殖与分化的影响。方法将兔成骨细胞分别与PDGF(10ng/ml),IGF-Ⅰ(100ng/ml),PDGF(10ng/ml)+IGF-Ⅰ(100ng/ml)共同培养,于1、3、5d分别进行3H-TdR、3H-Proline的掺入量以及碱性磷酸酶合成量的检测。结果PDGF与IGF-1联合应用对3H-TdR、3H-Proline的掺入量与单独使用因子组、对照组相比,在各个时段均存在显著性差异(P<0.01);PDGF与IGF-1联合应用对碱性磷酸酶合成量与对照组、PDGF组相比有显著性差异(P<0.01),与IGF-1组相比,未见明显差异(P>0.05)。结论PDGF与IGF-Ⅰ联合应用可显著促进成骨细胞DNA、胶原蛋白、碱性磷酸酶的合成,对促进成骨细胞的增殖与分化功能具有协同作用。
Objective To investigate the effects of PDGF combination with IGF-1 on proliferation and differentiation of cultured rabbit osteoblasts. Meth ods The second subcultures were exposed to PDGF (10ng/ml), IGF-1 (100ng/ml), an d PDGF (10ng/ml) combination with IGF-1 (100ng/ml). The DNA and collagen protei n synthesis were determined by the incorporation of 3H-TdR, 3H-Proline of oste oblasts respectively at 1st, 3rd and 5th days. Alkaline phosphatase activity in the media collected at 1st, 3rd and 5th days of culture was determined with king -Armstrong technique. Results The incorporation of 3H-TdR and 3H-Proline of o steoblasts in PDGF combination with IGF-Ⅰ group was statistically different a t the same period (P< 0.01)from that in the control group, the PDGF group, and t he IGF-Ⅰ group. Alkaline phosphatase activity of PDGF combination with IGF- Ⅰ group in same period was statistically different (P< 0.01)from that in the c ontrol group and the PDGF group, but it was not statistically different (P >0.05 ) from that of the IGF-1group. Conclusions The combination PDGF with IGF-Ⅰca n significantly stimulate the synthesis of DNA, collagen protein and alkaline ph osphatase of osteoblasts, and has a promoting effect on proliferation and differ entiation of osteoblasts.
