摘要
采用RAPD技术 ,对 2 8份玉米自交系的亲缘关系进行分析。旨在DNA水平上揭示玉米自交系之间的亲缘关系 ,为进一步提高玉米杂种优势利用水平提供有益的信息。从 10 0个 10bp随机引物中筛选出 2 4个多态性较好的引物 ,对 2 8份玉米自交系DNA进行扩增 ,扩增出 2 4张DNA指纹图谱 ,其中多态性DNA谱带 10 6条 ,占总扩增带数的 6 4 %。利用DNA扩增结果进行聚类分析 ,建立了 2 8个玉米自交系的亲缘关系树状图 ,将供试材料划分为五个类群 ,RAPD分析结果与已知系谱的亲缘关系基本一致。
This research was designed to identify the phylogenetic relationship of maize inbred using random amplified polymorphic DNA markers(RAPD).24 primers were selected from 100 arbitrary 10-mer primers to analyze 28 maize inbred.166 DNA bands were amplified with the 24 primers,106 of which (64%) were polymorphism.According to the RAPD data,a tree diagram of phylogenetic relationship was constructed using UPGMA cluster method in NYSYS.The results showed that 28 maize inbred could be divided into 5 groups at 0.78 similarity scale.It agrees with the taxonomic classification.The phylogenetic relationship of 28 maize inbred was clearly identified from the research.
